Gargi Mehta Lab started: 9/14/04 TLC Analysis of Analgesic Drugs Lab turned in: 10/05/04 Purpose: Thin layer chromatography was used to determine the composition of various analgesics and to identify the components in addition to the unknown analgesics. Apparatus: Chemicals needed: Chemicals L/S Amt 0.5% glacial acetic acid L small amt Absolute ethanol L 15 ml Methylene chloride L 15 ml Acetaminophen S small amount Aspirin S small amount Caffeine S small amount Ibuprofen S small amount Naproxen S small amount Tylenol tablet (drug of choice) S 500 mg Procedure: 1) Lightly draw a line using only lead pencil and not pen 1cm from the bottom of each reference plate. 2) On reference plate 1, starting from left to right, 3 component compounds were spotted (one drop only) in alphabetical order [Acetaminophen, Aspirin, Caffeine]. a. CAUTION – handle plates carefully or adsorbent may flake off and also handle by edges to avoid touching the surface. b. NOTE – spots should be made small as possible since to much of the sample may result in spots with tails overlapping. 3) On reference plate 2, starting from left to right, 2 component compounds were spotted (one drop only) in alphabetical order [Ibuprofen and Naproxen]. 4) On reference plate 3, starting from left to right, 2 component compounds were spotted (one drop only) [Instructors unknown and drug of choice]. 5) Each reference plate (one at a time) was then developed in a chamber containing 0.5% glacial acetic acid in ethyl acetate. 6) After plates were removed the solvent front position was marked using a lead pencil.
